We talked briefly about this at the workshop:

It might be nice to increase the lower and upper limits for the sedimentation coefficient on the frontend for the 2DSA 'fit control' box.

right now the lower limit is maxed at '10' and the upper limit at '100.'
I routinely have samples above 100, yet not very spread in s, so it might be helpful to set a range at say: 200 to 275 for example.

For me personally, I can imagine gold samples of up to 1000 Svedberg.

The lower and upper limit should both accommodate ranges between -10,000 to +10,000 s, front and backend. I have successfully measured ~ 10k s before. Once you get to about 1000 s though you have to run really slow which causes sometimes rotor stability issues, sedimentation by gravity alone, and ill-defined menisci. 3000 rpm is tolerable on the XLA. There could be some logic to make sure the lower limit is less than the upper limit. -0.1 through 0.1 s should be excluded from the range.

Mods have been made to frontend 2DSA and to LIMS to accept s values in the -10000 to 10000 range. The backend 2DSA computations should support this - with a skip of values in the -1e-14 to 1e-14 range (-0.1 to 0.1 before scaling by 1e-13) - on BCF and ALAMO as of July 20, 2011.

It is possible that backend computational code also already accepts the larger s range on other clusters. Once BCF and ALAMO have had sufficient testing, the other clusters will be upgraded.

Borries will run tests on data with higher sedimentation coefficient values (>200???) to verify that 2DSA can handle such values.
